AB We elaborated non-invasive non-infectious test-system suitable for
detection of anti-HIV antibodies in urine. We used at first synthetic
ELISA test-system Peptoscreen-2 (Institute for immunology, Moscow). But
if it is used according instruction for preparation it can not
differentiate intact urine from HIV-infected and non-infected persons.
To reach satisfactory results we invented two modifications. The 1-st is
method of very simple concentration of urine immunoglobulins by
self-salts-precipitation at freezing/thawing of intact urine. The 2-nd
is an introduction of new peptide antigen on solid phase. After
freezing/thawing a pellet is dissolved in 1/100 volume of PBS to initial
volume of fresh morning urine. The sensitivity of reaction is raised
approximately two-fold after absorption on solid phase the peptide aa.
603-624 (gp41) instead of peptide aa. 601-616. In urine we find anti-HIV
antibodies of IgG and IgA, but not IgM class. In some patients
concentration of anti-virus antibodies in such preparation from urine
was higher than in serum.
